Profile: Botanicals
For centuries, people have used herbs and other botanicals for their healthful effects, possibly due to their phytochemical composition, and in recent years, the use of these products has experienced an atmospheric rise in popularity among consumers. This has resulted in corresponding regulatory initiatives and the accelerated need for analytical methods for botanical products. Several regulatory initiatives, including proposed changes to current Good Manufacturing Practices (cGMP), address the issue of appropriate testing requirements for botanical ingredients.

Monograph Testing
Monograph testing is conducted using compendial methods such as the USP, FCC, and European, Chinese, British, and Japanese pharmacopeias (or other available pharmacopoeia) to ensure identity, strength, quality, purity, or potency. Following is a listing of some of the capabilities of the Covance monograph service:

Pricing for this testing is on an individual test basis; please inquire about pricing per individual compound and test. Standard analysis turnaround time is 8-15 days depending on analysis. Expedited service is available if all of the resources are available to schedule testing. When requesting monograph testing, please include compound general name, source of monograph test, specific test(s) needed, as well as requested turnaround time to ensure a timely quotation for this service.

Metallic Ion Species
Speciation (HPLC/ICP-MS)

Many elements occur at different valence states, forms, or compounds that can be detected using HPLC/ICP-MS analysis. The potential toxicity of different species can vary greatly. Evaluating the presence and concentration of each species will provide additional information that will aid in determining a compound's toxicity, bioavailability or mobility in a system of interest. The following table summarizes the species of the elements listed that could be separated and quantitated by HPLC/ICP-MS at Covance. The analysis is applicable to matrices not requiring decomposition such as water, physiological fluids, food additives, and extractable foods.

Consideration for performing speciated analysis is to obtain additional information that will aid in determining a compound's toxicity. The combination of HPLC with ICP-MS provides a rugged and sensitive system for speciation analysis. Sample preservation and preparation are critical for maintaining the species of interest until analysis. For example, toxic hexavalent chromium reduces to the trivalent form (an essential nutrient) in the presence of acid. Although speciation analysis is becoming more routinely applied, it is by no means mature. Additional information and pricing is available upon request.
